The victim, who has not yet been identified by police, suffered multiple gunshot wounds at Avalon club on Shaftesbury Avenue.
The Metropolitan Police were called at 3am and have closed the road – one of the main thoroughfares through the centre of the capital – while they investigate.
A man has been arrested and is being questioned at a south London police station, officers said.
It is not clear how long Shaftesbury Avenue will be closed, although bargain hunters are already descending on the area as shops re-open for the Boxing Day sales.
